In vitro effects of deltamethrin, pirimiphos-methyl and gamma-HCH on proteases in insecticide-resistant and susceptible strains of Tribolium castaneum

Abstract

To elucidate the possible involvement or the process of intraccllular protein catabolism in the development of insecticide resistance, we have compared the activities of a comprehensive range of cytoplasmic and lysosomal proteolytic enzymes in resistant and susceptible strains of red flour beetle, Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae). Compared to susceptible strain, the homogenate of adult beetles of resistant strain, showed higher activities of alanyl aminopeptidase (104%), arginyl aminopeptidase (205%), leucyl aminopeptidase (82%), dipeptidyl aminopeptidase IV (31%), tripeptidyl aminopeptidase (18%), proline endopeptidase (44%), dipeptidyl aminopeptidase I (158%), dipeptidyl aminopeptidase II (173%), cathepsin B (124%), Cathepsin H (199%) and Cathepsin D (25%). The only exception was Cathepsin L, which was decreased by 54%. Deltamethrin, pirimiphosmethyl and gamma-hexachlorocyclohexane (gamma-HCH), administered in vitro at a dose of 10-3M, resulted in elevation of only arginyl aminopeptidase (29%) and tripeptidyl aminopeptidase (57%) in resistant strain whereas gamma-HCH caused 22% increase of alanyl aminopeptidase in the susceptible strain. On the other hand, the activity of leucyl aminopeptidase was decreased by all the three insecticides. A dose of 10-6M of the insecticides did not affect any protease except leucyl aminopeptidase which showed depleted levels. The results suggested that induction of proteases in resistant strains may be of importance in the development of insecticide resistance.
